I am currently subscribed to a bunch of different topics that, when I logged in today, I noticed that people had replied to. However, I received no email notification, and I used to for other topics. I've had the experience where if someone posts in a thread I'm subscribed to while I'm online at astahost, I don't get emails (although I would prefer if I did, but I think that's a setting I can change), but when I logged off last night (read closed firefox) I was still the latest poster in some of the threads. So someone posted while I was offline, and I didn't get an email notification. I did get notification for some other threads though, so I know I have my settings right.
